Microdosing psilocybin or LSD means working with an amount intended to sit below a full psychedelic experience, often as part of a repeating practice woven into everyday life.

The reasons are personal: a brighter mood, renewed curiosity, deeper connection, more creative energy or a different relationship with discomfort. Research is beginning to give those experiences shape.

Brain basics

Psychedelics and the brain, in plain English.

Think of it as keys, locks and ripple effects. LSD and psilocin—the active form your body makes from psilocybin—are shaped enough like serotonin to fit some of the same locks in the brain.

01The molecule

Psilocybin becomes psilocin.

Your body converts psilocybin into psilocin. Psilocin and LSD do not simply add more serotonin; their shape lets them attach to some of the receptors normally used by serotonin.

02The receptor

The main lock is called 5-HT2A.

A receptor is like a lock on a brain cell. Both substances produce much of their psychedelic effect by turning 5-HT2A locks, especially on cells in the cortex—the outer layer involved in perception and thought.

03The ripple

That changes the conversation.

Those cortical cells change how they fire and influence glutamate, the brain’s main ‘go’ messenger. At larger research doses, familiar brain networks become less synchronized, allowing information to combine in less usual ways.

04Plasticity

The brain may become more change-ready.

Animal studies show that psilocybin can grow and reshape tiny connection points between neurons. Scientists call this plasticity. It is a window for change—not proof that every change is beneficial.

Psilocybin + mood

A month-long study found small-to-medium improvements in mood and mental health among psilocybin microdosers compared with non-microdosing participants, across age, gender and the presence of mental health concerns.

Psilocybin + the brain

In a double-blind human study, a psilocybin microdose produced a measurable reduction in theta-band EEG power. Preclinical research also found strengthened connections between the cortex and thalamus after repeated low doses.

A month moving in a brighter direction.

Researchers followed 953 psilocybin microdosers and 180 non-microdosing participants for about a month. The microdosing group showed small-to-medium improvements in mood and mental health, seen across age, gender and the presence of mental health concerns.

A large real-world comparison; encouraging, although it cannot prove that psilocybin caused the change.

A microdose left a measurable signature in brain rhythms.

In 34 participants, researchers compared psilocybin mushroom microdoses with placebo while measuring brain activity. The active condition reduced theta-band EEG power, a subtler version of the broader rhythm changes seen at higher doses.

A direct human neuroscience finding. The study did not find improved wellbeing, creativity or cognition, showing that a neural change is not automatically a functional benefit.

Repeated low doses strengthened a cortical–thalamic pathway.

In rats, a repeated low-dose psilocybin regimen increased resilience to stress, reduced compulsive actions and strengthened cortical connections to the paraventricular thalamic nucleus, a region involved in stress and emotional regulation.

A promising mechanistic clue rather than evidence of a human benefit; animal findings need to be tested in people.

Across six weeks, 80 healthy men received LSD or placebo. Daily reports showed improvements in creativity, connectedness, energy, happiness and wellness on LSD dose days, even after researchers accounted for participants’ expectations.

The shifts were short-term: overall mood and cognition did not show enduring change at six weeks.

Reading the evidence

Hopeful and honest can coexist.

    Observed improvement is a valuable signal. A randomized placebo-controlled result gives stronger evidence that the substance contributed to it.

    A positive dose-day experience is different from a lasting therapeutic change. Both can be meaningful, but they answer different questions.

  3. 03

    A measurable brain change is not automatically a therapeutic benefit. The human psilocybin EEG study found altered rhythms without improved cognition or wellbeing.

    These studies investigated specific research samples and settings. They do not establish microdosing as a treatment for a medical condition.
